The human studies I've read show that you can increase blood NAD levels with oral precursor supplementation (PMID 35479740), but IV use only appears to do so after at least 2 hours of infusion (PMID 31572171). Human studies have not shown significant supplementation and exercise performance benefits in young, healthy participants. Muscle NAD levels did not increase. Performance data in cohorts of elderly patients is mixed. Also, when looking at glucose and lipid metabolism, NAD precursors improved lipid levels but increased blood glucose. When analyzed by subgroups, there was little to no beneficial effect on healthy individuals, but there was an improvement in lipids with people with cardiovascular disease and abnormal lipids (PMID 3530395). My hypothesis with NAD was that supplementation likely was less effective than another well-documented modality to increase NAD: routine exercise. Routine exercise has already been proven to improve cardiometabolic markers and longevity. NAD levels do decline as we age, but this paper examined the available evidence and concluded that routine exercise protects against age-related NAD level decline and that the effect of exercise is greater than that of NAD precursor supplementation. I do not take NAD supplements, and the benefits are greatly exaggerated based on where human research is today, and the harms are downplayed.
