Duchenne muscular dystrophy (DMD) is a rare X-linked recessive genetic muscle disorder caused by a mutation in the dystrophin gene, leading to complete loss of the dystrophin protein — the key structure that protects muscle cells from damage. Without dystrophin, muscle fibers tear during contractions, causing progressive muscle weakness. Symptoms usually appear in early childhood: repeated falls, tiptoe walking, calf hypertrophy, Gowers sign (using hands to rise from the floor), bent knees, protruding abdomen, and scoliosis. Weakness starts in the proximal muscles and gradually affects the respiratory and cardiac muscles. By age 12, most patients require a wheelchair; by the late teens or early 20s, they need breathing and heart support. Sadly, life expectancy is often in the 20s or 30s. This video covers the cause, symptoms, inheritance, and disease progression of Duchenne muscular dystrophy — essential for medical students, healthcare learners, and anyone wanting to understand this inherited muscle disease.
