A chain of just 100 amino acids can, scientists estimate, fold into around 10 to the power 94 different shapes. If a computer checked one shape every trillionth of a second, it would still need about 10 to the power 75 years. The universe is only 13.8 billion years old. Yet real proteins fold in seconds. How? The chain doesn’t search randomly. Its own sequence guides it into one stable shape, and that shape decides what the protein does. Take amylase, the enzyme in your saliva. It folds so that a pocket forms in it, starch fits into that pocket like a key in a lock, and the enzyme starts breaking the starch into sugar. When folding goes wrong, things get dangerous. Boil an egg, and the heat unfolds the proteins in the white, so they clump together and turn solid. Cells can get it wrong too. In cystic fibrosis, a mutation makes a protein called CFTR misfold, and the cell destroys it before it reaches the surface. And it gets even crazier: in mad cow disease, a normal prion protein folds into the wrong shape. The amino acids stay the same, only the shape changes, and the misfolded protein can push others to misfold too. Same ingredients, different shape, different outcome. It’s like a 3D-printed part: same material, but get the shape slightly wrong and it won’t fit the machine. That’s why AI matters here. Google DeepMind’s AlphaFold predicts a protein’s final shape from its amino acid sequence alone. Its creators, Demis Hassabis and John Jumper, won the 2024 Nobel Prize in Chemistry for it. But it predicts the destination, not the journey, so how proteins fold, and misfold, is still being studied. Life works only when the shape is right.
